Altered Prefrontal-Amygdala Structural Connectivity in Adolescents Prenatally Exposed to Cocaine

چکیده
INTRODUCTION Prenatal cocaine exposure (PCE) is known to affect emotional regulation [1]. Reciprocal modulation between the ventral medial prefrontal cortex (VMPFC) and amgydala is associated with emotional control in healthy populations [2], and disconnection between is associated with behavioral dysregulation [3,4]. Previously our PCE cohort was shown to lack emotional suppression during cognitive demand; as compared to controls, the PCE group could not raise VMPFC activity during a higher cognitive load condition that contained negatively emotive distracters [5]. As these results imply a functional disconnect between the VMPFC and amygdala in the PCE group, and since VMPFC and amgydala are known to be structurally connected [6], the present study examined possible PCEinduced alterations to underlying structural connectivity.

Alterations in amygdala–prefrontal circuits in infants exposed to prenatal maternal depression
Prenatal exposure to maternal depression is common and puts offspring at risk for developing a range of neuropsychiatric disorders. Despite its prevalence and adverse associations, neurobiological processes by which prenatal maternal depression (PMD) confers risk remain poorly understood. Maternal mood and fetal behavior were assessed between 34 and 37 gestational weeks. Using resting-state fun...
متن کاملAltered amygdala connectivity in urban youth exposed to trauma.
Early life trauma exposure represents a potent risk factor for the development of mental illnesses such as anxiety, depression and post-traumatic stress disorder. Moreover, deleterious consequences of trauma are exacerbated in youth living in impoverished, urban environments. A priori probability maps were used to examine resting-state functional connectivity (FC) of the amygdala in 21 trauma-e...
متن کاملDysfunctional amygdala activation and connectivity with the prefrontal cortex in current cocaine users.
OBJECTIVES Stimulant use is associated with increased anxiety and a single administration of dexamphetamine increases amygdala activation to biologically salient stimuli in healthy individuals. Here, we investigate how current cocaine use affects amygdala activity and amygdala connectivity with the prefrontal cortex in response to biologically salient stimuli in an emotional face matching task ...
متن کاملAltered age-related trajectories of amygdala-prefrontal circuitry in adolescents at clinical high risk for psychosis: a preliminary study.
Emotion processing deficits are prominent in schizophrenia and exist prior to the onset of overt psychosis. However, developmental trajectories of neural circuitry subserving emotion regulation and the role that they may play in illness onset have not yet been examined in patients at risk for psychosis. The present study employed a cross-sectional analysis to examine age-related functional acti...

Prenatal cocaine exposure (PCE) is associated with long-term and negative effect on arousal regulation. Recent neuroimaging studies have examined brain mechanisms related to arousal dysregulation with cross-sectional experimental designs; but longitudinal changes in the brain, reflecting group differences in neurodevelopment, have never been directly examined. To directly assess the interaction...
